About the Position
We are seeking a highly motivated and skilled Laboratory Technician to join our research group focused on yeast functional genomics. Our lab has recently identified a large set of previously unannotated, potentially novel genes in Saccharomyces cerevisiae using deep RNA-seq and Ribo-seq experiments. The successful candidate will play a central role in experimentally characterizing these genes by generating and validating gene knockout mutants across multiple yeast strains.
Main Responsibilities
• Construct gene deletion mutants in S. cerevisiae using homologous recombination–based gene disruption methods.
• Design and assemble DNA constructs (PCR, cloning, primer design).
• Perform yeast transformations and confirm successful knockouts (PCR, sequencing, phenotypic verification).
• Maintain strain collections, plasmid stocks, and accurate laboratory records.
• Carry out growth assays under a range of stress conditions (e.g., temperature, osmotic, oxidative, nutrient deprivation) to assess the functional relevance of the targeted genes.
• Analyze growth data to identify phenotypic effects linked to gene deletions.
Required Qualifications
• Bachelor’s or Master’s degree in Molecular Biology, Genetics, Biotechnology, or a related field.
• Hands-on experience with Saccharomyces cerevisiae genetics or microbial model organisms.
• Strong skills in molecular techniques such as PCR, cloning, gel electrophoresis, and DNA preparation.
• Ability to work independently, manage multiple constructs/strains in parallel, and maintain detailed lab documentation.
Preferred Qualifications
• Experience with homologous recombination–mediated gene deletions in yeast.
• Experience performing growth assays or phenotyping under stress conditions.
• Familiarity with next-generation sequencing workflows or genomic analysis.
• Interest in gene function, genome annotation, or non-canonical translation.
